This study suggests the allele nomenclature of 14 cat STR markers according to the recommendations of the International Society for Forensic Genetics (ISFG) for human DNA typing, and it presents a standardized system for a secure DNA typing of samples.
Abstract
This is important for DNA typing in forensic casework, because the domestic cat is a popular pet, and owners usually have a close relationship with their cats. The alleles presented in this study are representative for a German cat population. It is possible that further alleles might occur by testing cats from other populations, but it can be expected that the majority of the described alleles in this study will be found in other populations of different areas. The recommendations of the ISFG state that allelic ladders of an STR marker should consist of the most common alleles appearing in a population studied. The allelic ladders compiled in this study consist of common as well as rare alleles. Only a small spectrum of samples has thus far been examined. It is possible that rarely occurring alleles can reach a higher frequency with an increased sample number and can then be included in the category of common alleles. Also, new alleles may arise that have not previously appeared. Due to the accurate investigation of 14 specific STR markers for the cat and their optimization in length and amplification, this report presents a good basic set for further forensic casework examinations. A Multiplex approach of the STR markers presented is being prepared, so that the combination of the presented STR markers in one or more sets also reduces the amount of DNA needed for PCR amplification. The nomenclature proposed and the standardization throughout allelic ladders confers advantages. 14 figures, 1 table, and 28 references
