This study validated use of the GlobalFiler Express PCR Amplification Kit for the direct amplification of reference DNA samples.
Abstract
The GlobalFiler Express PCR Amplification Kit is designed specifically for processing reference DNA samples in a high throughput manner. The kit uses 6-dye fluorescent chemistry to enable multiplexing of 21 autosomal STRs (short tandem repeats), 1 Y-STR, 1 Y-indel, and the sex-determining marker amelogenin. This article describes the studies that were conducted in order to validate use of the kit for the direct amplification of reference DNA samples in a high throughput manner. Blood and buccal samples from 10 individuals were collected and analyzed using the GlobalFiler system. The following types of assessments were conducted on the various samples: amplification; capillary electrophoresis and data analysis; cycle sensitivity study; reproducibility and precision study; a minimum threshold calculation and contamination assessment; and a concordance check. The results from these studies, highlighted in the article, suggest that the GlobalFiler Express PCR Amplification Kit produces reliable, robust, and reproducible results when use for amplifying blood and buccal reference DNA samples on punchable media. 3 tables, 3 figures, and 18 references
