To evaluate the usefulness of ABO genotyping by PCR (polymerase chain reaction) in paternity testing, blood samples were obtained from 80 unrelated Japanese and 13 disputed paternity cases.
Abstract
PCR amplification of DNA extracted from blood by a conventional extraction procedure was carried out using primers described in previous research. Amplified DNA products using four different primers were digested and then analyzed by electrophoresis on polyacrylamide gels. Bands on gels stained with ethidium bromide were observed with ultraviolet light and silver staining. In 20 samples of phenotype A, genotype AA was recognized in five samples; the other samples were genotype BO. According to the procedure, genotypes of phenotype AB and O were clearly and consistently confirmed. In 13 disputed paternity cases, the paternity of six cases was excluded with red cell, serum protein, enzyme genetic marker, and HLA (human leukocyte antigen) systems except ABO phenotype. The paternity of three of the six cases, however, was excluded with ABO genotype. Study findings appear to support the usefulness of ABO genotyping by PCR in paternity testing. 16 references, 1 table, and 2 figures
